Palmitoyl-CoA is a 16-carbon fatty acyl-CoA that serves as the main palmitate donor for protein N-palmitoylation and regulates mitochondrial function.¹ ² In development, it is the substrate for Hedgehog acyltransferase (Hhat), which palmitoylates Sonic hedgehog (Shh) and supports proper signaling through its transport into the ER lumen.² Elevated palmitoyl-CoA is also associated with lipotoxicity, mitochondrial dysfunction, and oxidative stress in metabolic disease.¹
Applications
Palmitoyl-CoA is used to study Hedgehog (Shh) signaling and Hhat function in developmental biology and cancer research, including drug targeting of this pathway (e.g., TDI-3410).² In metabolic research, it serves as a model intermediate for lipotoxicity, mitochondrial dysfunction, ROS production, apoptosis, and cardiolipin remodeling in diseases such as NAFLD.¹
