Lauroyl-CoA is a 12-carbon acyl-CoA that functions as a primer in mycolic acid biosynthesis in Mycobacterium tuberculosis, where it is utilized by β-ketoacyl-ACP synthase III (FabH). Structural studies show that it binds within an elongated hydrophobic channel, enabling transfer of the acyl group to the enzyme’s active-site cysteine. Lauroyl Coenzyme A also acts as a substrate for FabH in type II fatty acid synthesis (FASII) systems in bacteria and plants.¹
Applications
Lauroyl-CoA is used as a substrate in peroxisomal fatty acyl-CoA oxidase assays, supporting studies of peroxisomal β-oxidation and enzyme regulation.² It is also used in structural and biochemical studies of mycobacterial FabH, where it serves as a primer substrate for investigating bacterial fatty acid biosynthesis and guiding antimicrobial inhibitor development.¹ Lauroyl-CoA is further relevant to acyl-CoA thioesterase assays and investigations of enzyme chain-length specificity.³ ⁴ It may also be used in mitochondrial acyl-CoA dehydrogenase studies, depending on the enzyme and assay conditions, and might serve as an HPLC or LC-MS reference standard.⁵
