Myristoyl-CoA is a 14-carbon saturated acyl-CoA that functions as the essential donor for protein N-myristoylation. It is the preferred substrate for N-myristoyltransferases (NMTs), which attach myristate to the N-terminal glycine of target proteins.¹ ² Myristoyl-CoA exhibits very high binding affinity, enabling it to outcompete other acyl-CoAs.¹ The attached myristoyl group regulates membrane targeting, protein stability, and signal transduction.¹ ²

 

Applications

 

Myristoyl-CoA is used in drug discovery as a target for developing antifungal, antiviral, and anticancer inhibitors of N-myristoyltransferases.² Its high binding affinity also makes it a useful tool in proteomics for identifying substrate proteins, including LRATD1 and ERICH5. In enzymology, Myristoyl Coenzyme A is applied to study NMT kinetics and ordered Bi-Bi mechanisms, often using nonhydrolyzable analogs.¹ ²
