Stearoyl-CoA (C18:0-CoA) is a long-chain saturated fatty acyl-CoA that functions as a key metabolic intermediate in mammalian lipid biosynthesis.¹ ² It is generated from palmitoyl-CoA via fatty acyl-CoA elongase (Elovl6), under transcriptional control of SREBP-1, and serves as a central substrate in lipid metabolic pathways.¹

 

Applications

 

Stearoyl-CoA primarily serves as the substrate for stearoyl-CoA desaturase (SCD), which converts it into oleoyl-CoA, a key precursor for triglycerides, cholesterol esters, phospholipids, and wax esters. It also contributes to physiological lipid production in tissues such as skin and the eye, supporting lubrication and barrier function. In metabolism, stearoyl-CoA regulates energy balance by inhibiting acetyl-CoA carboxylase, lowering malonyl-CoA levels and promoting mitochondrial fatty acid oxidation. Clinically, the oleate-to-stearate ratio (desaturation index) is used as a biomarker of SCD activity and lipid metabolic status.²
