4′,4′′-Diphosphopantethine, also known as Pantethine 4′,4″-diphosphate, is a phosphorylated derivative of pantethine that functions as the preferred substrate for a NADH-linked disulfide reductase in Bacillus species. It is involved in the reduction of oxidized coenzyme A (CoA disulfides) back to active thiol forms, a function that is particularly important in bacteria lacking glutathione. In dormant spores, most CoA exists in disulfide form, and this reductase rapidly restores the active pool during early germination using NADH.¹
