4′-Phosphopantetheine (P-PantSH) is a central intermediate in coenzyme A (CoA) metabolism, linking its biosynthesis and degradation.¹ Unlike CoA, it can cross cell membranes without transporters and remains stable in circulation.¹ ² It supports mitochondrial function by activating mitochondrial acyl carrier protein and can restore metabolic balance in CoA-deficient states.² P-PantSH also exhibits antioxidant activity and participates in host–microbe interactions, as some bacteria secrete or depend on it for growth.¹ ³
Applications
4′-Phosphopantetheine has important roles in medicine, microbiology, and drug development.¹ ² ³ ⁴ P-PantSH is a promising therapeutic for pantothenate kinase-associated neurodegeneration (PKAN), where it bypasses defective CoA biosynthesis and restores mitochondrial function.¹ ² In cardiovascular research, it acts as an antioxidant that reduces ROS formation and atherosclerotic plaque development, and may serve as a biomarker for coronary disease progression.³ Its ability to cross membranes also makes it a useful tool for studying CoA metabolism and host–microbe interactions.¹ Additionally, enzymes in its pathway are targets for antimicrobial development, particularly against Staphylococcus aureus.⁴
