Octanoyl Coenzyme A (Octanoyl-CoA) is a medium-chain thioester intermediate used in microbial biosynthesis of high-value esters for flavor, fragrance, and biofuel applications.¹ ² In Saccharomyces cerevisiae, the enzyme Eht1 shows highest activity with C8 substrates and functions as both an acyltransferase and thioesterase, which may help regulate intracellular CoA levels during fermentation.² In Escherichia coli, octanoyl-CoA is a β-oxidation intermediate whose availability can be increased by deleting fadE to prevent degradation.¹

 

Applications

 

Octanoyl-CoA is used as a substrate in microbial production of esters such as ethyl octanoate and butyl octanoate.¹ ² It is applied in biochemical assays to study enzymes like Eht1, including their dual acyltransferase and thioesterase activity and substrate specificity for C8 chains.² In metabolic engineering, it is used to optimize ester yields by limiting β-oxidation and improving pathway flux.¹ Octanoyl coenzyme A also serves as a tool in protein engineering to enhance enzyme selectivity and enable production of single, non-toxic ester products in host systems such as E. coli
