Myristoyl Coenzyme A, Triammonium salt
CAS No.: 799812-85-4
Synonym(s): Myristoyl Coenzyme A, Triammonium salt, n-Tetradecanoyl Coenzyme A Triammonium salt
Myristoyl-CoA is a 14-carbon saturated acyl-CoA that functions as the essential donor for protein N-myristoylation. It is the preferred substrate for N-myristoyltransferases (NMTs), which attach myristate to the N-terminal glycine of target proteins.¹ ² Myristoyl-CoA exhibits very high binding affinity, enabling it to outcompete other acyl-CoAs.¹ The attached myristoyl group regulates membrane targeting, protein stability, and signal transduction.¹ ²
Applications
Myristoyl-CoA is used in drug discovery to investigate N-myristoyltransferase (NMT) as a target for antifungal, antiviral, and anticancer inhibitors.¹ ² Its high binding affinity also supports substrate-specificity studies and the identification of NMT substrate proteins, including LRATD1 and ERICH5.¹ In enzymology, myristoyl-CoA is used to study NMT kinetics, ordered Bi-Bi mechanisms, and substrate recognition, including through nonhydrolyzable analogs.² ³ It is also used in assays evaluating antifungal NMT inhibitors and in studies of protein myristoylation in Candida albicans.⁴ Myristoyl-CoA further supports research into GPI-anchor myristoylation in African trypanosomes.⁵ Myristoyl-CoA might be used as an HPLC or LC-MS reference standard.⁶
