Propionyl Coenzyme A (Propionyl-CoA) is a three-carbon acyl-CoA that functions as a central metabolic intermediate in both prokaryotic and eukaryotic organisms.¹ ² ³ It participates in multiple interconnected catabolic pathways, including the 2-methylcitric acid cycle, the acrylate pathway, and the methylmalonyl-CoA mutase pathway.¹ ³ In mammals, it is formed during the degradation of cholesterol side chains, odd-chain fatty acids, and amino acids such as valine, isoleucine, methionine, and threonine, while in bacteria it can be synthesized via enzymes such as PrpE or engineered routes from 2-ketobutyrate.¹ ² ³

 

Applications

 

Propionyl-CoA is widely used in metabolic engineering as a precursor for bioplastic production, particularly for generating the 3-hydroxyvalerate (3HV) monomer in poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)], a material with improved flexibility and toughness.² In medical research, it serves as a key metabolic marker in hepatocellular carcinoma, where reduced propionyl-CoA and propionyl-L-carnitine levels correlate with tumor progression and poor prognosis. Restored propionyl-CoA metabolism can suppress TCA cycle activity and reduce cancer cell proliferation through inhibition of citrate synthase.³
