5′-Iodo-2′,5′-dideoxy-thymidine 3′-CE phosphoramidite is used in DNA nanotechnology and synthetic biology to enable nonenzymatic ligation of oligonucleotides, allowing the assembly of large DNA constructs, circular DNA, and non-natural architectures without reliance on ligase enzymes. The incorporated 5′-iodide is stable under standard DNA synthesis and deprotection conditions and is introduced with high coupling efficiency.¹
